Florence will receive roughly $12 million in bonus money as part of his new contract. The Panthers signed Colclough to a two-year, $4 million deal.
Florence was considered one of the top free-agent cornerbacks despite having lost his starting spot to emerging star Antonio Cromartie after the 10th game last season.
Florence, a five-year veteran, was productive as a nickel corner in the playoffs. He intercepted two passes and defended four others.
The Jaguars chose not to re-sign Terry Cousin, the nickel back the last three years, and safety Sammy Knight. Both 11-year veterans turn 33 before next season.
Florence could fill Cousin's role or replace cornerback Brian Williams, who would then move to safety.
Florence played 74 games for the Chargers and started 43 of them. He has nine career interceptions and 47 passes defended.
"This is a move that will significantly strengthen our secondary," Jaguars coach Jack Del Rio said. "Drayton is entering the prime of his career and he's a real good fit for us and our defense."
